اثر دما بر سینتیک استخراج ترکیبات فنولی از ریزوم زنجبیل (Zingiber officinale) به کمک حمام فراصوت
در پژوهش حاضر تاثیر دماها (°C60-40) و زمانهای مختف ( min60-5) بر سینتیک استخراج ترکیبات فنولی و فلاونوئیدی ریزوم زنجبیل تحت شرایط حمام فراصوت بررسی و مقادیر IC50 آزمون مهار رادیکال DPPH و قدرت احیاکنندگی عصارهها محاسبه شد. نتایج نشان داد با افزایش دما، بازده استخراج ترکیبات فنولی و فلاونوئیدی افزایش یافت. بیشترین بازده این ترکیبات در min 15 اول دوره استخراج بهدست آمد و بعد از آن شیب استخراج کاهش یافت. همچنین بیشترین مقدار ترکیبات فنولی (69/47±75/165و µg GA/g of DW of extract56/36± 13/171) و فلاونوئیدی (80/0±43/9 و µg QE/g of DW of extract 93/0± 35/10) در انتهای مرحله استخراج سریع حدود min15، در دماهای 50 و °C60 بدون تفاوت معنیدار به دست آمد. بنابراین، فرآیند استخراج را میتوان به دو مرحله استخراج سریع و استخراج آهسته تقسیم کرد. نتایج نشان داد، با استفاده از نمودارهای سینتیک میتوان بیشینه استخراج ترکیبات آنتیاکسیدانی ریزوم زنجبیل را با کمترین زمان و دمای موثر بهدست آورد. علاوه بر این نتایج نشان داد روش فراصوت دارای کارایی قابل ملاحظهای در استخراج ترکیبات زیستفعال از ریزوم زنجبیل و تکنولوژی موثرتری نسبت به روش سنتی است.

الکتروریزپوشانی لیکوپن در ساختار میکروالیاف پروتئینی: بررسی خصوصیات فیزیکوشیمیایی و زیستدسترسی
لیکوپن رنگدانه کاروتنوئیدی چربیدوستی است که دارای خواص سلامتی بخش ویژهای میباشد، به این دلیل بسیاری از محققین، آن را به عنوان یک ماده ارزشمند میشناسند. پایداری بسیاری از ترکیبات زیست فعال به علت فرآیندهای گوناگون فیزیکوشیمیایی و فیزیولوژی محدود میشود، به همین منظور استفاده از روشهای مختلف ریزپوشانی جهت آزاد شدن کنترلشده و افزایش میزان پایداری ترکیبات زیست فعال، امری ضروری میباشد. در این پژوهش تأثیر غلظتهای مختلف محلول زئین (35 و 30، 25، 20، 15 درصد وزنی/حجمی) بر تولید الیاف زئین با استفاده از روش الکتروریسی مورد بررسی قرار گرفت. در ادامه از غلظت بهینه جهت ریزپوشانی لیکوپن در دو سطح (0.075، 0.05درصد وزنی/ وزنی) استفاده شده است. خصوصیات فیزیکی و شیمیایی الیاف زئین حاوی لیکوپن مورد بررسی قرار گرفت. همچنین پروفایل رهایش لیکوپن در سه فاز دستگاه گوارش انسان (دهان، معده و روده کوچک) بررسی شد. تصاویر حاصل از میکروسکوپ الکترونی روبشی (SEM) نشان داد الیاف یکنواخت، همگن و بدون مهره برای زئین حاصل گردیده است. راندمان ریزپوشانی لیکوپن در دو نمونه بین 88.07-85.68درصد متغیر بوده است. نتایج حاصل آزمون FTIR حاکی از آن است که ریزپوشانی بهصورت موفقیتآمیز صورت گرفته و از نوع فیزیکی میباشد. علاوه بر این رهایش کنترلشده و پایدار از لیکوپن در دستگاه گوارش مشاهده شده است که این سبب افزایش در زیستدسترسی آن شده است. ریزپوشانی با استفاده از الیاف الکتروریسی شده زئین پتانسیل بالایی به عنوان یک سیستم رسانش هدفمند مناسب برای لیکوپن دارد. از اینرو استفاده از این روش برای ریزپوشانی لیکوپن در صنعت غذا توصیه میشود.

پیش تیمار اکوتیپ ایرانی ماریتیغال با امواج مایکروویو و تاثیر آن بر کیفیت روغن استخراجی از دانه قلعه بابک
کاربرد تکنولوژیهای نوین از جمله پیشتیمار با امواج مایکروویو در دانههای روغنی منجر به افزایش راندمان روغن، مواد مغذی- دارویی و نیز پایداری اکسیداتیو بهتر روغن این دانهها میشود. در این پژوهش، دانههای ماریتیغال (اکوتیپ قلعه بابک، آذربایجان شرقی) تحت پیش تیمار مایکروویو (800 وات) در دو زمان مختلف (2 و 4 دقیقه ) قرار گرفتند و تاثیر آن روی بهبود راندمان روغن استخراجی، ویژگیهای فیزیکوشیمیایی، پروفیل اسیدهای چرب و میزان توکوفرولها در روغن استخراجی از آن بررسی شد. برای مقایسه نتایج از روغن دانه ماریتیغال بدون تیماردهی با مایکروویو به عنوان نمونه کنترل استفاده شد. نتایج نشان داد که پیشتیمار مایکروویو دانه ماریتیغال راندمان روغن استخراج شده، مقدار فنل کل و توکوفرولها را در این نمونهها افزایش داد. همچنین برخی ویژگیهای فیزیکوشیمیایی روغن ماریتیغال شامل میزان کلروفیل (91/1-03/1 میلی گرم فئوفیتین درکیلوگرم روغن) و عدد صابونی ( 188-181 میلی گرم پتاس درگرم روغن) با تیمار مایکروویو افزایش یافتند، ولی عدد اسیدی (14/2-20/4 میلی گرم پتاس درگرم روغن)، شاخص پراکسید (23/3-22/6 میلی اکی والان اکسیژن درکیلوگرم روغن) و عدد یدی (100-109 گرم ید در100گرم روغن) با تیمار مایکروویو کاهش پیدا کردند. نتایج نشان داد که تاثیر پیش تیمار مایکروویو روی اسیدهای چرب روغن ماریتیغال ناچیز بود، بطوری که برخی از اسیدهای چرب از قبیل اسید اولئیک (C18:1) و اسید لینولئیک (C18:2)، با تیمار مایکروویو کاهش ولی اسید پالمیتیک (C16:0) و اسید استئاریک (C18:0) افزایش یافتند. همچنین نتایج نشان داد که با افزایش زمان تیماردهی با مایکروویو میزان اسیدهای چرب غیر اشباع بطور ناچیزی کاهش یافتند. در کل، نتایج نشان داد که پیش تیمار با مایکروویو منجر به افزایش میزان استخراج روغن و توکوفرول ها در روغن دانه ماریتیغال میشود.

بررسی آزمایشگاهی و مدلسازی سینتیک خشک کردن شلتوک در خشک کن هوای گرم توسط شبکه عصبی مصنوعی
برنج در هنگام برداشت رطوبت بالایی دارد که یکی از روش های رایج جهت کاهش سرعت فساد، فرآیند خشک کردن است. در این تحقیق سینتیک خشک کردن شلتوک با هوای گرم با استفاده از 3 متغیر سرعت هوای گرم ((m/s)65/0 و 8/0)، دمای هوای گرم ((0C)50،65) و رطوبت نهایی (11 و 13%) بررسی شد. مدلسازی سینتیک خشککردن شلتوک توسط شبکه عصبی پرسپترون چند لایه انجام شد. به منظور بررسی عملکرد الگوریتمهای آموزش و توابع انتقال مختلف در پیشبینی رفتار خشککردن شلتوک، از 3 الگوریتم لونبرگ مارکوارت، پس انتشار ارتجاعی و گرادیان نزولی مقیاسی و 2 تابع انتقال تانژانت سیگموئید و سیگموئید لگاریتمی استفاده شد. نتایج حاصل از این بررسی بدین گونه بوده که حداکثر زمان خشک شدن در دمای 0C50، سرعت هوای گرم (m/s)65/0 و رطوبت نهایی 11٪ تقریباً برابر با 8 ساعت و حداقل زمان خشک شدن در دمای 0C 65، سرعت هوای گرم(m/s) 8/0و رطوبت نهایی 13٪ معادل با 1ساعت و 21 دقیقه بوده است. بنابراین به طور کلی می توان گفت با افزایش دما، سرعت و رطوبت، زمان خشک شدن کاهش مییابد. همچنین، نتایج حاصل از مدل سازی نشان داد که مدلسازی بوسیلهی شبکه عصبی با الگوریتم آموزش لونبرگ مارکوارت نسبت به سایر الگوریتمها بهترین عملکرد را داشته و بهطورکلی توپولوژی 1-11-3 با الگوریتم آموزش لونبرگ مارکوارت و تابع انتقال سیگموئید لگاریتمی کمترین خطا و بیشترین ضریب همبستگی نسبت به سایر حالات داشته است.

طبقهبندی خواص بیولوژیکی میوه گلابی در بارگذاری دینامیکی و استاتیکی با استفاده از شبکه عصبی مصنوعی
شبکههای عصبی مصنوعی تکنیکهای مدلسازی قدرتمند هستند که با آرایههایی از نورونها در حافظه و یادگیری بیولوژیک کار میکنند. در این تحقیق به بررسی طبقهبندی نوع بارگذاری دینامیکی و شبه استاتیکی (لبه پهن و لبه نازک) با استفاده از دادههای ورودی محتوای فنول، انتی اکسیدان، ویتامین C و سفتی با شبکه عصبی مصنوعی پرداختهشده است. در این آزمایش برای طبقهبندی از دو شبکه تابع پایه شعاعی و پرسپترون چندلایه با دو تابع فعال سازی تانژانت هیپربولیک و سیگموئیدی در یکلایه مخفی با نرون هایی به تعداد 4 و 8 نرون استفاده شد. با توجه به نتایج بدست آمده بهترین مقدار R و درصد صحیح (Percent Correct) برای بارگذاری دینامیکی ( =100 – R=9999997درصد صحیح)، بارگذاری لبه نازک (=100 – R=9999993درصد صحیح) و بارگذاری لبه پهن (=100 – R=9999992درصد صحیح) بود، که در شبکه تابع پایه شعاعی با تابع فعالسازی سیگوئید و تعداد 8 نورون در لایه مخفی ایجادشده است. همچنین بیشترین دادههای صحیح تشخیص دادهشده برای نوع بارگذاری دینامیکی، لبه پهن و لبه نازک در شبکههای ایجادشده برای شبکه تابع پایه شعاعی مشاهده شد و این شبکه توانسته است که بهطور 100 درصد برای تمامی بارگذاریها میزان دادهها را بهدرستی طبقهبندی کند. درمجموع شبکه عصبی با ورودی دادههای کلی توانایی مطلوبی را در طبقهبندی دادهای بارگذاری دینامیکی و شبه استاتیکی داشته است.

بررسی تآثیر شرایط پیش تیمار اکستروژن بر خصوصیات فیزیکوشیمیایی مخلوط آرد بدون گلوتن برنج و ذرت
آرد برنج و ذرت جایگزین مناسبی برای توسعه محصولات بدون گلوتن هستند و اصلاح برخی خصوصیات آنها با فرایندهای مختلف میتواند در جهت بهبود کیفیت محصولات بدون گلوتن مؤثر باشد. این پژوهش با هدف بررسی تأثیر فرایند اکستروژن بر خصوصیات فیزیکوشیمیایی آرد بدون گلوتن مخلوط برنج و ذرت به نسبت 1:1 انجام شد. بدین منظور تیمارهای اکستروژن با متغیرهای دمای اکستروژن 110، 145 و °C180 و میزان رطوبت 12، 15 و 18 % روی مخلوط آرد بدون گلوتن ذرت و برنج اعمال گردید و ویژگیهای شاخص جذب آب و حلالیت، نشاسته صدمه دیده، مؤلفههای رنگی، دانسیته توده و خصوصیات حرارتی توسط آزمون کالریمتری روبشی تفاضلی یا DSC بررسی شد. نتایج نشان داد که اعمال فرایند اکستروژن سبب تغییرات معنیداری (05/0P<) در ویژگیهای مورد بررسی شده است، بهطوری که شاخص جذب آب، حلالیت و نشاسته صدمه دیده از 88/1 g/g ، 5/4 % و 8 UCD در نمونه شاهد به 1/7- 4/4 g/g و 5/28 – 4/11 % و 8/28-2/20 UCD در نمونههای مورد تیمار افزایش پیدا کرد درحالیکه دانسیته توده و میزان روشنایی آرد بهترتیب به حداقل 48/0 و 65/75 کاهش یافتند. بررسی خصوصیات حرارتی حاکی از آن بود که دماهای ژلاتیناسیون شامل دمای شروع یا To، دمای پیک یا Tp و دمای خاتمه یا Tc بهطور معنیداری افزایش یافتند ولی آنتالپی ژلاتیناسیون یا ∆E از j/g 93/7 در نمونه شاهد به دامنه j/g 41/5- 08/1 در نمونههای مورد تیمار کاهش پیدا نمود. در نهایت با توجه به تغییرات رخ داده در نمونههای آرد طی فرایند اکستروژن، میتوان نتیجه گرفت که این فرایند پیش تیمار مناسبی برای تولید آردهایی با خصوصیات عملکردی متفاوت میتواند باشد و در تهیه محصولات مختلف بدون گلوتن مفید واقع گردد.

تاثیر حمام التراسونیک، نسبت سورفاکتانت به روغن و غلظت موسیلاژ دانه به بر ویژگیهای نانوامولسیون خودبخودی
هدف از این مطالعه بررسی ویژگیهای نانوامولسیون روغن در آب تثبیت شده با مخلوط ایزوله پروتئین آب پنیری (1 درصد وزنی/حجمی) و موسیلاژ دانه به (1/0 و 5/0 درصد وزنی/حجمی) به روش کم انرژی خودبخودی و پر انرژی هموژنایزر فراصوت است. ازآنجاکه مشکل اصلی روش امولسیون سازی خودبخودی استفاده از مقادیر بالای سورفاکتانتهای سنتزی است بنابراین یکی از اهداف این تحقیق بررسی تأثیر نسبتهای مختلف سورفاکتانت به روغن (1:1، 2:1 و 3:2 =SOR) و تیمار کمکی حمام التراسونیک روی ویژگیهای نانوامولسیون خودبخودی بود. پارامترهای اندازه ذرات، پتانسیل زتا، ویسکوزیته ظاهری و پایداری نانوامولسیون مورد بررسی قرار گرفت. در این مطالعه امولسیونهای با قطر ذرات کمتر از 200 نانومتر بهخوبی تهیه شدند. افزایش غلظت موسیلاژ و نسبت سورفاکتانت به روغن باعث کاهش معنیدار اندازه ذرات، افزایش اندک پتانسیل زتا (افزایش نگاتیویتی)، افزایش ویسکوزیته، کاهش اندیس خامهای شدن و افزایش شاخص پایداری امولسیونها شد. نمونههای حاصل از روش پر انرژی و نمونههای که با حمام التراسونیک تیمار شدهاند دارای کمترین اندازه ذرات، کمترین مقدار شاخص خامهای شدن و بیشترین شاخص پایداری امولسیون بودند. نتایج این تحقیق نشان داد که میتوان با یک تیمار ساده توسط حمام التراسونیک میتوان نانوامولسیون خودبخودی با ذرات بسیار کوچک (در ابعاد نانومتر) بدون نیاز به غلظتهای بالای سورفاکتانتهای سنتزی را تهیه کرد.

تأثیر شرایط فرایند و فرمولاسیون بر ویژگیهای فیزیکوشیمیایی و عملکردی مکمل فیبری بافت داده شده (تفالۀ گوجهفرنگی-سبوس برنج)
صنعتغذا سالانه حجم عظیمی از پسماند را در سرتاسر جهان تولید مینماید این ترکیبات منبع ارزشمندی پروتئینها، فیبر رژیمی، آنتیاکسیدانها و انواع ریز مغذیها میباشند از این رو مدیریت کاربرد مجدد پسماند حاصل از فرآوری نهادههای کشاورزی با حداکثر راندمان، به منظور تولید فراوردههایی با ارزش افزوده و ارزان قیمت، از اهمیت ویژه ای برخوردار است. اخیراً فرآوری انواع پسماند توسط تکنولوژی پخت اکستروژن به عنوان فرایندی با کارایی بالا مورد توجه قرار گرفته است از این رو در پژوهش حاضر، بر پایۀ طرح مرکب مرکزی چرخش پذیر اثر متغیرهای فرمولاسیون شامل نسبت مختلف تفالۀ گوجهفرنگی : سبوس برنج ( 25:75- 75:25 درصد وزنی: وزنی)، میزان رطوبت خوراک (18-12 درصد) و شرایط فرایند اکستروژن شامل سرعت چرخش مارپیچ (160-120 دور بر دقیقه) بر ویژگیهای عملکردی و فیزیکوشیمیایی مکمل فیبری اکسترود شده (تفالۀ گوجهفرنگی - سبوس برنج) شامل شاخص جذب روغن، دانسیتۀ توده، مؤلفه رنگی(روشنایی، قرمزی و زردی) ، فعالیت آنتیاکسیدانی و فعالیت آبی بررسی گردید. نتایج نشان داد ویژگیهای فراوردۀ نهایی متأثر از فرمولاسیون خوراک و شرایط اکستروژن بود، بهطوریکه افزایش سرعت چرخش مارپیچ، سبب افزایش شاخص جذب روغن فراورده گردید افزایش نسبت تفالۀ گوجهفرنگی به سبوس برنج به طور مستقل موجب افزایش مؤلفههای رنگی (قرمزی و زردی) و کاهش میزان روشنایی (شاخصL*) و دانسیتۀ توده شد افزایش همزمان دو پارامتر رطوبت و سرعت چرخش مارپیچ باعث کاهش فعالیت آنتی اکسیدانی گردید و فعالیت آبی نمونهها با افزایش همزمان رطوبت و نسبت مکملهای فیبری، افزایش یافت. مطابق نتایج بهینهیابی به منظور ارائه مکمل فیبری اصلاح شده برای کاربرد در فراوردههای غذایی رژیمی دانسیتۀ توده 6/0گرم بر سانتیمتر مکعب، فعالیت آبی 17/0، شاخص جذب روغن60/2گرم بر گرم، فعالیت آنتی اکسیدانی14/35درصد و میزان روشنایی 18/57، شرایط فرایند شامل میزان رطوبت خوراک 26/13درصد، نسبت تفالۀ گوجهفرنگی به سبوس برنج 25:75 درصد وزنی: وزنی و سرعت چرخش مارپیچ 63/137دور بر دقیقه، تعیین گردید.

بررسی تأثیر عصاره اتانولی بره موم بر ویژگیهای فیزیکوشیمیایی، ریزساختاری، آنتی اکسیدانی و ضدمیکروبی فیلم مرکب نشاسته – ژلاتین – پلی وینیل الکل
در این مطالعه، فیلم زیست تخریب پذیر مرکب بر پایه نشاسته گندم، ژلاتین و پلی وینیل الکل تهیه شد و از عصاره اتانولی بره موم (5، 10 و 20 %) به منظور ایجاد خواص ضدمیکروبی و آنتی اکسیدانی در آن استفاده شد. فیلمها به روش قالب ریزی حلال تهیه شدند و خواص ریزساختاری، مکانیکی، بازدارندگی، ظاهری، آنتی اکسیدانی و ضدمیکروبی آنها مورد ارزیابی قرار گرفت. در نمونه شاهد بدلیل عدم سازگاری بیوپلیمرها با یکدیگر، فیلم یکنواختی تشکیل نشد اما با افزودن عصاره، یکنواختی فیلم افزایش یافت و مطابق تصاویر SEM، میزان ترکها و شکافها در فیلم کمتر شد. آزمون FT-IR تشکیل اتصالات جدید بین بیوپلیمرها و ترکیبات عصاره بره موم را تأیید نمود. افزودن عصاره باعث کاهش استحکام کششی، نفوذ پذیری بخار آب و کدورت فیلم مرکب گردید اما ازدیاد طول تا نقطه شکست و اندیس زردی را افزایش داد. محتوای فنل کل و ظرفیت آنتی اکسیدانی فیلم مرکب با افزایش غلظت عصاره افزایش یافت. غلظتهای مختلف عصاره بر روی تمام ویژگیهای فیلم مرکب به جز میزان جذب رطوبت اثر معنیدار داشت. فیلمهای حاوی عصاره اثرات ضد میکروبی قویتری بر روی باکتری S.aureus در مقایسه با باکتری E.coli از خود نشان دادند. بهطور کلی فیلم حاوی 10 درصد عصاره بهترین ویژگیهای فیزیکوشیمیایی و عملکردی را نشان داد. نتایج این پژوهش نشان داد که فیلم مرکب حاوی عصاره بره موم میتواند به عنوان بسته بندی آنتی اکسیدانی و ضدمیکروبی به منظور افزایش ماندگاری مواد غذایی مورد استفاده قرار گیرد.

بررسی اثر نوع آنزیم و زمان هیدرولیز برتولید پپتیدهای آنتی اکسیدان از پروتئین اسپیرولینا پلاتنسیس
شرایط هیدرولیز آنزیمی پروتئین ها شامل نوع آنزیم، زمان هیدرولیز و درجه هیدرولیز بر فعالیت زیستی محصول هیدرولیز پروتئین تاثیر گذار است. هدف از تحقیق حاضر، بررسی اثر شرایط هیدرولیز آنزیمی بوسیله آنزیم های پپسین ،کیموتریپسین و ترکیب دو آنزیم (نسبت آنزیم به سوبسترای 1:10، زمان 5 ساعت، دمای 37 درجه سانتیگراد) بر میزان تولید پپتیدهای آنتی اکسیدان از پروتئین ریزجلبک اسپیرولینا پلاتنسیس استخراج شده با روش سونیکاسیون (200 وات ، 20 کیلوهرتز، زمان 5 دقیقه) بود. میزان پیشرفت هیدرولیز آنزیمی با روش(OPA) O-phethaldialdehyde و فعالیت آنتی اکسیدانی محصول هیدرولیز پروتئین، بر اساس دو مکانیسم مهارکنندگی رادیکالهای DPPH و ABTS در طی زمان هیدرولیز بررسی شدند. مقدار گروههای آمین آزاد بعد از 5 ساعت هیدرولیز از مقدار 272/3 میکرومول لوسین/میلی گرم پروتئین به مقادیر 653/3 و 522/4 و 713/3 به ترتیب برای آنزیم های پپسین، کیموتریپسین و ترکیب دو آنزیم رسید. فعالیت آنتی اکسیدانی محصول هیدرولیز پروتئینی در طی زمان هیدرولیز افزایش یافت. بطوریکه فعالیت مهارکنندگی رادیکال های DPPH و ABTS بعد از 150 دقیقه هیدرولیز بوسیله پپسین به ترتیب به مقادیر 23 و 110 میکرومول ترولوکس/ میلی گرم پروتئین رسید. درحالیکه هیدرولیز بوسیله آنزیم کیموتریپسین فعالیت آنتی اکسیدانی را بعد از 120 دقیقه به ترتیب به مقادیر 18 و 159 میکرومول ترولوکس/میلی گرم پروتئین رساند و ترکیب دو انزیم باعث افزایش فعالیت مهار کنندگی DPPH تا مقدار 25 میکرومول ترولوکس/ میلی گرم پروتئین (بعد از 90 دقیقه) و مهارکنندگی ABTS تا مقدار 230 میکرومول ترولوکس/ میلی گرم پروتئین (بعد از 210 دقیقه) شد بطور کلی نتایج نشان دادند که هیدرولیز پروتئین ریزجلبک اسپیرولینا پلاتنسیس بوسیله آنزیمهای پپسین و کیموتریپسین و ترکیب دو آنزیم باعث تولید پپتیدهای آنتی اکسیدان می شود و آنزیم کیموتریپسین آنزیم موثرتری از پپسین و استفاده از ترکیب دو آنزیم پپسین و کیموتریپسین موثرتر از استفاده تکی از آنزیم هاست.
